Antibody Immunogen Description: Corresponds to amino acid residues R974 to M980. The effect of Y977 phosphorylation is unclear. This is the major site of in vivo phosphorylation of PLCG1 (≥665 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is conserved in mammals and found in the rat.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PLCG1 (1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase gamma-1) mediates the production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). It plays an important role in the regulation of intracellular signaling cascades, regulating crucial cellular processes including for growth, proliferation, migration, and apoptosis. It becomes activated in response to ligand-mediated activation of receptor-type tyrosine kinases, such as PDGFRA, PDGFRB, FGFR1, FGFR2, FGFR3 and FGFR4. It also functions in actin reorganization. It acts as a calcium-dependent phospholipase C, binding to calcium ions and regulatory proteins.
