Antibody Immunogen Description: Corresponds to amino acid residues R1093 to E1103. The effect of S1096 and T1100 phosphorylation is unclear. These are two of the major in vivo phosphorylation sites in PLCE1 (≥19 and >5 reports, respectively, by mass spectrometry). These human phosphosites are conserved in mammals and found in the rat.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PLCE1 (Phospholipase C epsilon-1) is a 230 kDa bifunctional enzyme that acts as both a lipase, generating DAG and IP3 to regulate cell signaling, and a Guanine Nucleotide Exchange Factor (GEF) for Ras family proteins. It activates the Ras/MAPK signaling pathway and controls Ca2+ signaling, impacting cell survival and actin organization. It features an N-terminal RasGEF domain, central catalytic X and Y domains, and two C-terminal Ras-associating (RA) domains. Unlike other PLCs, it lacks a pleckstrin homology domain. PLCE1 is a bifunctional enzyme which also regulates small GTPases of the Ras superfamily through its Ras guanine-exchange factor (RasGEF) activity. It regulates cell growth, differentiation, and motility, primarily in the heart, kidney, and digestive tract.
