Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PLCB3

Antibody Immunogen Sequence: RHN(pS)ISEAC

Antibody Immunogen Description: Corresponds to amino acid residues R1102 to A1109. S1105 phosphorylation inhibits enzyme activity. This is the major in vivo phosphorylation site in PLCB3 (≥232 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is conserved in mammals and also found in the rat. PLCB3 is known to be phosphorylated at this site in vitro by PKACa (PRKACA), PKCa (PRKCA), PKCb1 (PRKCB1), PKCd (PRKCD), PKCe (PRKCE), PKG1 (PRKG1), and PKG2 (PRKG2).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Scientific Background: PLCB3 (Phospholipase C-β3) is an essential effector enzyme that mediates G protein-coupled receptor (GPCR) signaling by catalyzing the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2} into secondary messengers diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). It is activated by G-alpha-q, G-alpha -11, and G-beta/gamma subunits. It regulates crucial cellular processes like calcium release, protein kinase C (PKC) activation, and trans-Golgi network transport. PLCB3 is a key signaling enzyme that initiates intracellular, receptor-mediated transduction in response to hormones and neurotransmitters. It is particularly critical for mediating inflammatory responses and cellular signaling. PLCB3 plays a significant role in inflammation, particularly in the lungs. It acts on the membrane surface, where its activity is regulated by a "plug" domain that, when removed by membrane binding, allows for interaction with activated G proteins. It features a pleckstrin homology (PH) domain that mediates binding to complexed G-beta/gamma subunits. Its activity is further controlled by calmodulin and it can form homodimers. Mutations or loss of function in PLCB3 are associated with Spondylometaphyseal Dysplasia with Corneal Dystrophy (SMDCD), a disorder characterized by growth deficiencies, skeletal abnormalities, and eye issues.
