Antibody Immunogen Description: Corresponds to amino acid residues R383 to G389. The effect of S386 phosphorylation is unclear. This is the major in vivo phosphorylation site in PFKP (≥234 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the fruit fly.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PFKP (ATP-dependent 6-phosphofructokinase, platelet type) is a key regulatory 85 kDa homotetramic enzyme that belongs to the phosphofructokinase type A (PFKA) family, ATP-dependent PFK group I subfamily. It catalyzes the phosphorylation of D-fructose 6-phosphate to fructose 1,6-bisphosphate by ATP, the first committing step of glycolysis. It is highly expressed in platelets, fibroblasts, and various cancer cells, where it promotes proliferation, migration, and survival. While primarily in the cytoplasm, it also operates at the mitochondria, lysosomal membrane, and nucleus. Its activity is enhanced by factors such as c-Myc and AKT, supporting tumour growth. It plays a central role in the Warburg effect (aerobic glycolysis) in cancer, with high expression in breast, lung, and brain tumours.
