Antibody Immunogen Description: Corresponds to amino acid residues M514 to I521. S518 phosphorylation inhibits activity. Phosphorylation induces binding of Ezrin, Merlin and SPTAN1, and inhibits interaction with CD44, DDB1, Hrs, LIN28B, LRP6, Merlin, PIK3R2, SRc, TUBA4A and VPRBP. This is the major in vivo phosphorylation site in NF2 (≥37 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in fish. NF2 is known to be phosphorylated at this site in vitro by PAK1, PAK2, and PKACa (PRKACA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NF2 (Moesin-ezrin-radixin-like protein; Merlin; schwannomin) is a tumour suppressor protein that belongs to the ERM (Ezrin, Radixin, Moesin) protein family. It links membrane proteins to the cytoskeleton. It regulates cell adhesion, motility, and contact-dependent inhibition of proliferation. It acts as a scaffold, regulating signaling pathways like Hippo/SWH and mTOR to control proliferation. The Hippo/SWH (Sav/Wts/Hpo) signaling pathway plays a pivotal role in tumour suppression by restricting proliferation and promoting apoptosis. Along with WWC1, it can synergistically induce the phosphorylation of LATS1 and LATS2. It may act as a membrane stabilizing protein. It can inhibit PI3 kinase by binding to AGAP2 and impairing its stimulating activity. It suppresses cell proliferation and tumorigenesis by inhibiting the CUL4A-RBX1-DDB1-VprBP/DCAF1 E3 ubiquitin-protein ligase complex.NF2 features a FERM domain and a C-terminal domain. It exists in "open" (active) and "closed" (inactive) conformations, often regulated by phosphorylation, such as at Ser-518. While widely expressed, it is primarily produced in the nervous system (especially Schwann cells). It is typically found in actin-rich areas like filopodia, lamellipodia, and cell boundaries. Isoform 1 and isoform 3 predominate. Isoform 4, isoform 5 and isoform 6 are expressed moderately. Isoform 8 is found at low frequency.
