Antibody Immunogen Description: Corresponds to amino acid residues G410 to N416. Y413 phosphorylation induces transcription. This is the major in vivo phosphorylation site in IKZF1 (≥94 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the chicken.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: IKZF1 (DNA-binding protein Ikaros) is a 58 kDa transcription regulator of hematopoietic cell differentiation that belongs to the Ikaros C2H2-type zinc-finger protein family. It features four N-terminal zinc fingers for DNA binding and two C-terminal zinc fingers for dimerization/interaction with other proteins. It binds gamma-satellite DNA, and activates the enhancer (delta-A element) of the CD3-delta gene. It plays a role in the development of lymphocytes, B- and T-cells, and increases normal apoptosis in adult erythroid cells. It acts as a repressor of the TDT (fikzfterminal deoxynucleotidyltransferase) gene during thymocyte differentiation. It regulates transcription through association with both HDAC-dependent and HDAC-independent complexes. It targets the 2 chromatin-remodeling complexes, NuRD and BAF (SWI/SNF), in a single complex (PYR complex), to the beta-globin locus in adult erythrocytes.
