Antibody Immunogen Description: Corresponds to amino acid residues R366 to S372. The effect of S369 phosphorylation is unclear. This is the major in vivo phosphorylation site in GATA3 (≥28 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the fruit fly. GATA3 is known to be phosphorylated at this site in vitro by Pim.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: GATA3 (Trans-acting T-cell-specific transcription factor GATA-3) is a transcriptional activator which binds to the enhancer of the T-cell receptor alpha and delta genes. It binds to the consensus sequence 5'-AGATAG-3'. It plays a vital role in the development of the mammary gland, urothelium, and immune system. It is required for the T-helper 2 (Th2) differentiation process following immune and inflammatory responses. It positively regulates ASB2 expression. GATA3 coordinates macrophage transcriptional activation and UCP2-dependent metabolic reprogramming in response to the cytokine IL33. In response to tissue injury, it acts downstream of IL33 signaling to drive differentiation of inflammation-resolving alternatively activated macrophages. It is expressed in T-cells and in luminal cells of the breast and the urinary tract. It is crucial for the development of multiple tissues, including the nervous system, kidneys, and skin. Mutations in the GATA3 gene are associated with HDR syndrome (Hypoparathyroidism, Sensorineural Deafness, and Renal Dysplasia). GATA3 is a highly sensitive diagnostic marker for luminal breast cancer (appearing in up to 94% of cases) and urothelial carcinoma.
