Antibody Immunogen Description: Corresponds to amino acid residues G204 to T210. The effect of S207 phosphorylation is unclear. This is one of the major in vivo phosphorylation sites in fatty acid synthetase (≥42 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the nematode worm.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: FASN (Fatty acid synthase) is a vital, homodimeric multi-enzyme protein (encoded by the FASN gene in humans) that catalyzes the de novo synthesis of long-chain fatty acids, primarily palmitate (C16:0), from acetyl-CoA, malonyl-CoA, and NADPH. It is a cytosolic, multifunctional enzyme with seven distinct catalytic activities, and a site for the binding of the prosthetic group 4'-phosphopantetheine of the acyl carrier protein ([ACP]) domain. It catalyzes the reaction: acetyl-CoA + n malonyl-CoA + 2n NADPH + 2n H+ = a long-chain fatty acid + (n+1) CoA + n CO2 + 2n NADP+. It is highly expressed in brain, lung, and liver.
