Antibody Immunogen Description: Corresponds to amino acid residues Q443 to F449. Y446 phosphorylation activates COX2 enzymatic activity. However, this phosphosite has been poorly detected in mass spectrometry studies (≥1 report by mass spectrometry). The phosphosite is commonly conserved in vertebrates and found in fish. COX2 is known to be phosphorylated at this site in vitro by Fyn.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: COX2 (Prostaglandin G/H synthase 2; PTGS2) is an inducible enzyme that belongs to the prostaglandin G/H synthase family. It catalyzes the conversion of arachidonate ((5Z,8Z,11Z,14Z)-eicosatetraenoate, AA, C20:4(n-6)) to prostanoids, a class of C20 oxylipins, including prostaglandin H2 (PGH2). The cyclooxygenase activity of COX2 oxygenates AA to the hydroperoxy endoperoxide prostaglandin G2 (PGG2), and its peroxidase activity reduces PGG2 to the hydroxy endoperoxide prostaglandin H2 (PGH2), the precursor of all 2-series prostaglandins and thromboxanes. It is constitutively expressed in some tissues in physiological conditions, such as the endothelium, kidney and brain, and in pathological conditions, such as in cancer. PTGS2 is responsible for production of inflammatory prostaglandins. Up-regulation of PTGS2 is also associated with increased cell adhesion, phenotypic changes, resistance to apoptosis and tumour angiogenesis. In cancer cells, PTGS2 is a key step in the production of prostaglandin E2 (PGE2), which plays important roles in modulating motility, proliferation and resistance to apoptosis.
