Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDKN2A (Cyclin-dependent kinase inhibitor 2A; p16-INK4A) belongs to the CDKN2 cyclin-dependent kinase inhibitor family. It acts as a negative regulator of the proliferation of normal cells by interacting strongly with CDK4 and CDK6. This inhibits their ability to interact with cyclins D and to phosphorylate the retinoblastoma protein (Rb), effectively blocking cell cycle progression from the G1 phase to the S phase. It is widely expressed but not detected in brain or skeletal muscle. Isoform 3 is pancreas-specific. Note that the gene that encodes CDKN2A also specifies the tumour suppressor ARF, which is distinct from CDKN2A. p19ARF binds to and promotes the degradation of MDM2, a molecule that typically breaks down the tumour suppressor p53. By inhibiting MDM2, p14ARF stabilizes p53, enabling it to arrest the cell cycle or trigger apoptosis (cell death). CDKN2A is a well-known susceptibility gene for familial atypical multiple mole melanoma syndrome. It is frequently mutated, deleted, or methylated in a variety of tumours, including non-small cell lung cancer, pancreatic cancer, and lymphoid malignancies.
