Antibody Immunogen Description: Corresponds to amino acid residues R506 to L512. T509 phosphorylation induces transcription. This is the major site of phosphorylation of BRCA1 by mass spectrometry studies (≥79 reports). This human phosphosite is not highly conserved in nature, but is found in the mouse. BRCA1 is known to be phosphorylated at this site in vitro by Akt1.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: BRCA1 (Breast cancer type 1 susceptibility protein) is a transcription protein, coactivator/corepressor and ubiquitin ligase. As a E3 ubiquitin-protein ligase that specifically mediates the formation of 'Lys-6'-linked polyubiquitin chains, it plays a central role in DNA repair by facilitating cellular responses to DNA damage. It iremains unclear whether it also mediates the formation of other types of polyubiquitin chains. The E3 ubiquitin-protein ligase activity is required for its tumour suppressor function. The BRCA1-BARD1 heterodimer coordinates a diverse range of cellular pathways such as DNA damage repair, ubiquitination and transcriptional regulation to maintain genomic stability. It regulates centrosomal microtubule nucleation, and is required for normal cell cycle progression from G2 to mitosis. It is also needed for appropriate cell cycle arrests after ionizing irradiation in both the S-phase and the G2 phase of the cell cycle. It is involved in transcriptional regulation of P21 in response to DNA damage, and required for FANCD2 targeting to sites of DNA damage. It may function as a transcriptional regulator. It inhibits lipid synthesis by binding to inactive phosphorylated ACACA and preventing its dephosphorylation. It contributes to homologous recombination repair (HRR) via its direct interaction with PALB2, fine-tunes recombinational repair partly through its modulatory role in the PALB2-dependent loading of BRCA2-RAD51 repair machinery at DNA breaks. BRCA1 is also a component of the BRCA1-RBBP8 complex which regulates CHEK1 activation and controls cell cycle G2/M checkpoints on DNA damage via BRCA1-mediated ubiquitination of RBBP8.
