Antibody Immunogen Description: Corresponds to amino acid residues E129 to D136. Y132 and Y133 phosphorylation stimulates phosphatase activity, and regulates cell adhesion, cell motility, and transcription. These are two major in vivo phosphorylation site in ACP1 (≥207 and >86 reports, respectively, by mass spectrometry in PhosphoSitePlus). These phosphosites are highly conserved and also found in yeast. ACP1 is known to be phosphorylated in vitro at Y132 by Lck, Src. Y133: Lck, and Src.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ACP1 (low molecular weight phosphotyrosine protein phosphatase; Acid phosphatase 1) is a widely expressed, 18 kDa enzyme that hydrolyzing protein tyrosine phosphate to protein tyrosine. In addition to dephosphorylating tyrosine phosphorylated proteins, it also targets low-MW aryl phosphates and natural and synthetic acyl phosphates with differences in substrate specificity between isoform 1 and isoform 2. It modulates key signaling molecules, including insulin and platelet-derived growth factor (PDGF) receptors. T-lymphocytes express only isoform 2. A third isoform does not possess phosphatase catalytic activity. It is primarily found in the cytosol and nucleoplasm.
