Antibody Immunogen Description: Corresponds to amino acid residues D679 to A686. Phosphorylation of Y682 increases enzyme activity. This is the most frequently phosphorylated site in ATP-citrate lysate in mass spectrometry studies (≥383 reports in PhsophositePlus). This human phosphosite is highly conserved in vertebrates and also found in the nematode worm. ACLY is known to be phosphorylated at this site in vitro by Lyn, Src.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ACLY (ATP-citrate synthetase) catalyzes the cleavage of citrate into oxaloacetate and acetyl-CoA with the reversible reaction: oxaloacetate + acetyl-CoA + ADP + phosphate <=> citrate + ATP + CoA. It is the primary enzyme responsible for synthesizing cytosolic acetyl-CoA, which is essential for de novo lipogenesis and cholesterogenesis. Acetyl-CoA is a common substrate in multiple biochemical reactions in protein, carbohydrate and lipid metabolism. As a homotetramer, it links glucose metabolism to lipid biosynthesis and histone acetylation. It features an N-terminal citrate-binding domain (Rossmann fold), a CoA binding domain, and a C-terminal citrate synthase domain. Found in the cytosol and nucleus of cells, it is highly active in the liver and adipose tissue during carbohydrate feeding.
