Antibody Immunogen Description: Corresponds to amino acid residues P249 to Q256. The effect of S253 phosphorylation is unclear. This is one of the major sites of in vivo phosphorylation of THRAP3 (≥148 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). THRAP3 is known to be phosphorylated at this site in vitro by ERK1 (MAPK3).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: THRAP3 (Thyroid Hormone Receptor Associated Protein 3; TRAP150) is a ubiquitous 150-kDa nuclear protein that belongs to the BCLAF1/THRAP3 family. It serves as a key transcriptional coactivator and regulator of pre-mRNA splicing, particularly within nuclear speckles, where it is important for maintaining the structure of the nucleolus. It plays essential roles in the DNA damage response, R-loop resolution, and adipocyte differentiation. As a transcriptional coactivation, it interacts with the thyroid hormone receptor (TR) and is involved in mediating gene expression. It functions in RNA processing as a component of the spliceosome, to regulate alternative splicing and mRNA stability. It remains associated with spliced mRNA after splicing which probably involves interactions with the exon junction complex (EJC). It can trigger mRNA decay which seems to be independent of nonsense-mediated decay involving premature stop codons (PTC) recognition.
