Antibody Immunogen Description: Corresponds to amino acid residues S367 to H375. T371 phosphoryation generates a docking site for Plk1 at S435. This is one of the major sites of in vivo phosphorylation of TERF1. This is a major site of in vivo phosphorylation of TERF1 based on ≥9 mass spectrometry reports recorded in PhosphoSitePlus. TERF1 is known to be phosphorylated at this site in vitro by CDK1 (CDC2).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TERF1 (Telomeric Repeat-Binding Factor 1; TRF1) is a homodimeric component of the shelterin complex (telosome) that binds to double-stranded 5'-TTAGGG-3' telomeric repeats. As part of the shelterin complex, it stabilizes telomeres and regulates access of telomerase. It acts as a negative regulator of telomerase, controlling telomere length by inhibiting elongation and preventing end-to-end fusions. TERF1 protects chromosome ends from degradation and is essential for proper telomere replication and mitotic spindle assembly. Shelterin associates with arrays of double-stranded 5'-TTAGGG-3' repeats added by telomerase and protects chromosome ends; without its protective activity, telomeres are no longer hidden from the DNA damage surveillance and chromosome ends are inappropriately processed by DNA repair pathways. It is present at telomeres throughout the entire cell cycle. It features a C-terminal Myb DNA-binding motif and an N-terminal dimerization domain (TRFH domain).
