Antibody Immunogen Description: Corresponds to amino acid residues G638 to T645. Y641 phosphorylation stimulates transcription and induces DNA and STAT6. This is one of the major sites of in vivo phosphorylation of STAT6 based on ≥96 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT6 (Signal transducer and activator of transcription 6) is a STAT family transcription factor that primarily mediates interleukin 3/ IL-3 signaling and interleukin-4 (IL-4) and plays a critical role in differentiation of T helper 2 (Th2) cells, Th2 immune responses, allergic inflammation, and B-cell immunoglobulin E (IgE) isotype switching. Like other STAT transcription factors, it features an SH2 domain for receptor association and a DNA-binding domain for transcription. Upon tyrosine phosphorylation by JAK's after their activation by IL3 and IL4 receptors, it dimerizes, translocates to the nucleus, and activates target gene expression.
