Antibody Immunogen Description: Corresponds to amino acid residues D776 to G784. S780 phosphorylation regulates molecular association, including inhibiting interaction with ERK1. This is a minor site of in vivo phosphorylation of STAT5A (≥25 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). STAT5A is known to be phosphorylated at this site in vitro by CDK8, ERK1 (MAPK3), ERK2 (MAPK1), and PAK1.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT5A (Signal transducer and activator of transcription 5A) is a STAT family transcription factor.that acts as a signaling molecule and transcription activator, crucial for mediating responses to and growth factors by regulating gene expression, primarily promoting cell survival and differentiation. It mediates cellular responses to various cytokines (e.g., IL-2, IL-3, Prolactin) that bind to cytokine receptors, and to growth factors (KITLG/SCF, FGFR1, FGFR2, FGFR3 and FGFR4) that activate receptor-tyrosine kinases (ERBB4). Upon cytokine stimulation, STAT5A is phosphorylated by JAK's on Tyr- 694, forming homo- or heterodimers (typically with STAT5B) that translocate to the nucleus.
