Antibody Immunogen Description: Corresponds to amino acid residues G689 to V697. Y693 phosphorylation induces interaction with LIFR, STAT3 and STAT4. This is the major sites of in vivo phosphorylation of STAT4 based on ≥8 mass spectrometry recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT4 (Signal transducer and activator of transcription 4) is an 86 kDa, STAT family transcription factor that regulates gene expression in response to cytokines, crucial for immune cell maturation and function. It is primarily activated by IL-12, IL-23, and Type I IFNs, promoting T-helper 1 (Th1) cell differentiation. It is also essential for producing interferon-gamma (IFG-g), and mediating responses to viral/bacterial pathogens. Upon cytokine signaling, cytoplasmic STAT4 is phosphorylated, dimerizes, and translocates to the nucleus to bind specific DNA regions. It is inhibited by PIASx (protein inhibitor of activated STAT), which acts as a co-repressor. It is expressed primarily in the thymus, spleen, and peripheral blood leukocytes. Variations in the STAT4 gene are strongly associated with increased risk for autoimmune diseases, including Systemic Lupus Erythematosus (SLE), Rheumatoid Arthritis (RA), Sjögren’s syndrome, and type 1 diabetes.
