Antibody Immunogen Description: Corresponds to amino acid residues D723 to T730. S727 phosphorylation stimulates transcriptional activity and induces interaction with DNA, p300, and inhibits interaction with gp130. Phosphorylation regulates carcinogenesis, cell differentiation, cell growth, cell motility, and transcription. This is a major site of in vivo phosphorylation of STAT3 (≥122 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). STAT3 is known to be phosphorylated at this site in vitro by CDK1 (CDC2), CDK5, DYRK1A, DYRK2, ERK1 (MAPK3), ERK2 (MAPK1), HIPK2, IRAK1, JNK1 (MAPK8), JNK2 (MAPK9), MAPKAPK2, MEKK1 (MAP3K1), MSK1 (RPS6KA5), mTOR (FRAP1), NLK, p38a MAPK (MAPK14), PKCd (PRKCD), PKCe (PRKCE), RSK2 (RPS6KA3), and TBK1 (NAK).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT3 (Signal transducer and activator of transcription 3) is a transcription factor of the STAT family that mediates cellular responses to interleukins, KITLG/SCF, LEP and other growth factors. Once activated, it recruits coactivators, such as NCOA1 or MED1, to the promoter region of the target gene (PubMed:17344214). It mediates cellular responses to activated FGFR1, FGFR2, FGFR3 and FGFR4. It also binds to the interleukin-6 (IL-6)-responsive elements identified in the promoters of various acute-phase protein genes.
