Antibody Immunogen Description: Corresponds to amino acid residues E686 to R694. Y690 phosphorylation stimulates transcriptional activity. This has not been identified as an in vivo phosphorylation of STAT2 from high throughput mass spectrometry studies recorded in PhosphoSitePlus. STAT2 is known to be phosphorylated at this site in vitro by JAK1, Lck, and Tyk2.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT2 (Signal transducer and activator of transcription 2) is a transcription factor of the STAT family that mediates signaling by type I IFNs (IFN-alpha and IFN-beta). Following type I IFN binding to cell surface receptors, JAK kinases (TYK2 and JAK1) are activated and catalyze tyrosine phosphorylation of cytoplasmic STAT1 and STAT2. Unlike other STATs, STAT2 generally does not form functional homodimers. The phosphorylated STATs dimerize, associate with IRF9/ISGF3G to form a complex termed ISGF3 transcription factor, which enters the nucleus. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of interferon stimulated genes, which drive the cell in an antiviral state. It acts as a regulator of mitochondrial fission by modulating the phosphorylation of DNM1L at Ser-616 and Ser-637 which activate and inactivate the GTPase activity of DNM1L respectively. It is essential for the innate immune response to viruses (e.g., Influenza A, Hepatitis) by facilitating interferon-stimulated gene (ISG) transcription.
