Antibody Immunogen Description: Corresponds to amino acid residues G698 to E705. Y701 phosphorylation stimulates transcriptional activity and induces interaction with DNA, PIAS1 and STAT2. This is a major site of in vivo phosphorylation of STAT1 (≥65 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). STAT1 is known to be phosphorylated at this site in vitro by FGFR3, JAK1, JAK2, JAK3, NPM-ALK, Src, and Tyk2.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAT1 (Signal Transducer and Activator of Transcription 1-alpha/beta) is a critical protein that acts as a transcription factor, mediating cellular responses to interferons (IFN-a/b/g), cytokines (KITLG/SCF), and growth factors (FGF's that stimulate FGFR1, FGFR2, FGFR3 and FGFR4). STAT1 exists as two isoforms: the full-length 91 kDa STAT1-alpha and the shorter, splice variant STAT1-beta. It is essential for innate and adaptive immunity, controlling gene expression for antiviral, antibacterial, and antiproliferative responses. It is activated via phosphorylation at Tyr-701 and by JAK kinases (JAK1, TYK2) in response to signaling molecules. Upon phosphorylation, STAT1 forms homodimers (Gamma-activated factor - GAF) or heterodimers (with ISGF3G/IRF-9 to form ISGF3) that translocate to the nucleus to induce gene transcription. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of IFN-stimulated genes (ISG), which drive the cell in an antiviral state.
