Antibody Immunogen Description: Corresponds to amino acid residues V921 to L929. The effect of T925 phosphorylation is unclear. This is a major in vivo site for phosphorylation of SH2BP1 based on ≥126 mass spectrometry reports recorded in PhosphoSitePlus. SH2BP1 is known to be phosphorylated at this site in vitro by CK2a1 (CSNK2A1).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SH2BP1 (RNA polymerase-associated protein; CTR9 homolog) is a 150 kDa component of the PAF1 complex (PAF1C), which plays a central role in transcriptional regulation by RNA polymerase II. It is a large, nuclear protein (approximately 150 kDa) that acts as a scaffold for the assembly of the PAF1 complex and connects transcription to various histone modifications. The N-terminal two-thirds of the protein contains 15–16 tetratricopeptide repeats (TPRs), which mediate protein-protein interactions and self-association. As part of the PAF1C, SH2BP1 is involved in transcriptional elongation, acting in cooperation with other factors to facilitate RNA polymerase II movement along DNA. It is required for specific histone modifications, including the monoubiquitination of histone H2B (H2BK120ub1) and trimethylation of histone H3 Lys-4 (H3K4me3). It is also involved in regulating the distribution of H3K27me3, acting to counterbalance the Polycomb Repressive Complex 2 (PRC2). PAF1C is required for transcription of Hox and Wnt target genes. PAF1C is involved in hematopoiesis and stimulates transcriptional activity of KMT2A/MLL1.
