Antibody Immunogen Description: Corresponds to amino acid residues N257 to P264. S260 phosphorylation inhibits transcriptional activity, inhibits interaction with DNA, RARB, RXRA and VDR, and regulates transcription, intracellular location, and protein stability. This is a minor site of in vivo phosphorylation of RXRa based on ≥2 mass spectrometry reports recorded in PhosphoSitePlus. RXRa is known to be phosphorylated at this site in vitro by ERK1 (MAPK3), ERK2 (MAPK1), JNK1 (MAPK8), and JNK2 (MAPK9).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RXRA (Retinoid X Receptor alpha) is a ~55 kDa nuclear receptor and ligand-dependent transcription factor that belongs to the nuclear hormone receptor family, NR2 subfamily. It plays a critical role in regulating gene expression for cell proliferation and differentiation, apoptosis, lipid metabolism, and embryonic development. It binds to 9-cis retinoic acid and acts as a central dimer partner for various nuclear receptors (e.g., PPARs, RARs) to regulate metabolic and developmental pathways. It forms homo- or heterodimers to bind retinoic acid response elements (RAREs) in target gene promoters.The RAR/RXR heterodimers bind to RARE composed of tandem 5'-AGGTCA-3' sites known as DR1-DR5. In the absence of a ligand, RXR heterodimers bind corepressors. Ligand binding causes corepressor dissociation and coactivator recruitment, activating transcription.
