Antibody Immunogen Description: Corresponds to amino acid residues L31 to K38. S35 phosphorylation induces interaction with CTNNB1. This is a minor site of in vivo phosphorylation of Ron based on ≥1 high throughput mass spectrometry reports recorded in PhosphoSitePlus. RORA is known to be phosphorylated at this site in vitro by PKCa (PRKCA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RORA (RAR-related orphan receptor alpha) is a nuclear receptor transcription factor that belongs to the nuclear hormone receptor family, NR1 subfamily. Cholesterol and its derivatives (e.g., 25-hydroxycholesterol) act as natural ligands (agonists), while 7-oxygenated sterols act as inverse agonists. It acts as a monomer to regulate gene expression, crucial for cerebellar development, circadian rhythm, immune response (notably Th17 differentiation), and lipid/glucose metabolism. It binds to ROR response elements (RORE) containing a single core motif half-site (5'-AGGTCA-3'). RORA modulates genes involved in cerebellar development (e.g., SHH), photoreceptor development (OPN1SW, OPN1SM, ARR3), skeletal muscle development (MYOD1), circadian rhythm (BMAL1, CLOCK, NPAS2, CRY1) and lipid metabolism (APOA1, APOA5, APOC3, PPARG). RORA is essential for the development of type 2 innate lymphoid cells (ILC2) and promotes Th17 cell differentiation. In liver, it has specific and redundant functions with RORC as positive or negative modulator of expression of genes encoding phase I and phase II proteins involved in the metabolism of lipids, steroids and xenobiotics, such as CYP7B1 and SULT2A1. It induces a rhythmic expression of some of these genes. In addition, RORA interplays functionally with NR1H2 and NR1H3 for the regulation of genes involved in cholesterol metabolism. In adipose tissue, it functions as a negative regulator of adipocyte differentiation, probably acting through dual mechanisms.
