Antibody Immunogen Description: Corresponds to amino acid residues G85 to K95. The effects of S89 and T91 phosphorylation are unclear. These are two of the major sites of in vivo phosphorylation of RDBP based on ≥105 and ≥186 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RDBP (RD RNA-binding protein; NELFE) is a 45.4–45.6 kDa subunit of the five-component Negative Elongation Factor (NELF) complex. It belongs to the RRM NELF-E family. It plays a critical role in controlling gene expression by regulating RNA polymerase II (Pol II) transcript elongation. RDBP/NELFE works with DSIF (DRB sensitivity-inducing factor) to repress transcription by inducing Pol II pausing shortly after transcription begins. The NELF complex is counteracted by the P-TEFb kinase complex. It features a unique, long region of alternating basic and acidic residues, specifically arginine (R) and aspartic acid (D). While it is proposed to be a RNA-binding protein (RBP) and contains an RNA Recognition Motif (RRM) that binds to nascent TAR RNA to help stabilize the NELF complex at pause sites, its direct RNA-binding has been challenged. RDBP is expressed in heart, brain, lung, placenta, liver, skeletal muscle, kidney and pancreas. The NELF complex is involved in HIV-1, potentially recruiting factors to the paused polymerase.
