Antibody Immunogen Description: Corresponds to amino acid residues E388 to D393. S392 phosphorylation induces activity, stimulates binding to DNA, NFkB-p65, UBE4B and p21Cip1, and inhibits binding of CDK1 and MDM2. This is one of the minor in vivo phosphorylation sites in p53 (≥13 reports by mass spectrometry in PhosphositePlus), although there are ≥94 reports with Western blotting. TP53 is known to be phosphorylated at this site in vitro by ATM, CDK7, CDK9, CK2a1 (CSNK2A1), ERK1 (MAPK3), LKB1 (STK11), NuaK1, and PKR (EIF2AK2).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: The p53 protein ( tumour protein 53 or TP53) is a DNA-binding cell cycle-regulating transcription factor that governs cell division and the fine balance between cell death and cell survival (1). P53 plays a critical role in tumour suppression and hence it is often described as "the guardian of the genome", "the guardian angel gene", or the "master watchman." This also refers to its role in conserving stability by preventing genome mutation (2).
