Antibody Immunogen Description: Corresponds to amino acid residues L14 to W23. T18 and S20 phosphorylation stimulates binding of CBP/p300, and inhibits binding to its negative regulatory partner MDM2. T18 phosphorylation has not been reported by mass spectrometry studies, and phosphorylation of the S20 site has only been reported once by mass spectrometry, although ≥117 reports for S20 phosphorylation has been made based on Western blotting data. TP53 is known to be phosphorylated in vitro at T18 by AMPKA1, Chk1 (CHEK1), Chk2 (CHEK2), CK1a1 (CSNK1A1), CK1d1 (CSNK1D), DAPK1, DNAPK (PRKDC), VRK1, and VRK2, and at S20 by AMPKa1 (PRKAA1), ATM, CDK5, Chk1 (CHEK1), Chk2 (CHEK2), CK1a1 (CSNK1A1), CK1d1 (CSNK1D), DAPK1, DAPK3 (ZIPK), DNAPK (PRKDC), DRAK1 (STK17A), JNK1 (MAPK8), JNK2 (MAPK9), MAPKAPK2, and Plk3 (CNK). .

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
