Antibody Immunogen Description: Corresponds to amino acid residues K604 to K613. The effects of T607 and T610 phosphorylation are unclear. These are two of the major in vivo phosphorylation sites in NOLC1 (≥102 and ≥101 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). NOLC1 is known to be phosphorylated in vitro at T607 and T610 by CDK1 (CDC2).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NOLC1 (Nucleolar and Coiled-Body Phosphoprotein 1) is a 140 kDa chaperone of the NOLC1 family.It chaperones proteins between the nucleolus and cytoplasm, and plays a critical role in ribosome biogenesis, rRNA transcription, and neural crest development. It acts as a platform for RNA Polymerase I and is highly phosphorylated. It undergoes rapid, massive phosphorylation and dephosphorylation by CK2 and PKC. It has intrinsic GTPase and ATPase activities. It acts as a chaperone that links RNA Polymerase I with enzymes responsible for ribosomal processing. It regulates RNA polymerase I by interacting with the largest subunit (RPA194) and aids in processing small nucleolar ribonucleoproteins (snoRNPs). It is required for neural crest specification.
