Antibody Immunogen Description: Corresponds to amino acid residues S899 to S910. S903 phosphorylation promotes protein proteolysis. This is one of the major in vivo phosphorylation sites in NFKB1 (≥30 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). NFKB1 is known to be phosphorylated at this site in vitro by GSK3b.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NFkB1 (Nuclear Factor Kappa B Subunit 1) is a 105 kDa precursor protein (p105), which is proteolytically processed by the 26S proteasome to generate the p50 subunit, a crucial DNA-binding component of the NF-κB transcription factor complex. It plays a key role in immune response, inflammation, and cell survival, usually by forming a p50-p65 heterodimer. This protein contains an N-terminal Rel homology domain (RHD) for DNA binding/dimerization and a C-terminal region with ankyrin repeats. NFKB1/p50 has both repressive and activating roles in NF-κB transcription factor complex that regulates genes. NFkB consists of homo- or heterodimeric complexes formed from the five NFkB proteins in mammals: RelA, RelB, c-REL, NFkB1, and NFkB2. They form a variety of homodimers and heterodimers, each binding at kappa-B sites in the DNA, regulating distinct sets of genes.
