Antibody Immunogen Description: Corresponds to amino acid residues G210 to A220. S213 and S217 phosphorylation stimulates transcriptional activity. These are two of the major in vivo phosphorylation sites in NFAT3 (≥12 and ≥16 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). NFAT3 is known to be phosphorylated at this site in vitro by S213 and S217 by JNK1 (MAPK8), and JNK2 (MAPK9).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NFAT3 (Nuclear factor of activated T-cells, cytoplasmic 4) is a Ca(2+)-regulated transcription factor that binds the consensus DNA sequence 5'-GGAAAAT-3'. It is involved in several processes, including the development and function of the immune, cardiovascular, musculoskeletal, and nervous systems. It binds to PPARG and REG3G gene promoters. It is involved in T-cell activation, stimulating the transcription of cytokine genes, including for IL2 and IL4. Along with NFATC3, NFAT3 is involved in embryonic heart development. It is involved in mitochondrial energy metabolism required for cardiac morphogenesis and function. It transactivates many genes involved in the cardiovascular system, including AGTR2, NPPB/BNP (in synergy with GATA4), NPPA/ANP/ANF and MYH7/beta-MHC. It is involved in the regulation of adult hippocampal neurogenesis, BDNF-driven pro-survival signaling in hippocampal adult-born neurons, and the formation of long-term spatial memory and long-term potentiation. In cochlear nucleus neurons, NFAT3 may play a role in deafferentation-induced apoptosis during the developmental critical period, when auditory neurons depend on afferent input for survival. It binds to and activates the BACE1/Beta-secretase 1 promoter, and may regulate the proteolytic processing of the amyloid precursor protein (APP). It plays a role in adipocyte differentiation, and may be involved in myoblast differentiation into myotubes. In the presence of CREBBP, it activates TNF transcription. It is widely expressed, with high levels in placenta, lung, kidney, testis, ovary, and heart, with weaker expression in spleen and thymus.
