Antibody Immunogen Description: Corresponds to amino acid residues S863 to K871. S867 phosphorylation inhibits binding to ER-alpha. This is one of the less major in vivo phosphorylation sites in NCOA3 (≥12 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). NCOA3 is known to be phosphorylated at this site in vitro by CDK1 (CDC2), JNK1 (MAPK8), and p38a MAPK (MAPK14).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NCoA3 (Nuclear Receptor Coactivator 3; SRC-3; AIB) is a ~155-160 kDa transcription coactivator protein that belongs to the SRC/p160 nuclear receptor coactivator family. It stimulates the transcriptional activities of nuclear receptors in a hormone-dependent fashion. It plays a central role in creating a multisubunit coactivator complex, which probably acts via remodeling of chromatin. It is involved in the coactivation of different nuclear receptors, such as for steroids (GR and ER), retinoids (RARs and RXRs), thyroid hormone (TRs), vitamin D3 (VDR) and prostanoids (PPARs). It enhances nuclear receptor activity by recruiting CBP/p300 and PCAF to DNA, promoting gene expression through histone acetyltransferase activity. It is also involved in the coactivation of the NF-kappa-B pathway via its interaction with the NFKB1 subunit. It is widely expressed, with highest expression in heart, skeletal muscle, pancreas and placenta, lower expression in brain, and very low levels in lung, liver and kidney. It is often overexpressed in cancers, and it acts as an oncogene promoting cell cycle progression and therapy resistance.
