Antibody Immunogen Description: Corresponds to amino acid residues P176 to Q183. S180 phosphorylation stimulates transcriptional activity. This is the major in vivo phosphorylation site in MEF2D (≥51 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). MEF2D is known to be phosphorylated at this site in vitro by ERK5 (MAPK7).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MEF2D (Myocyte-specific enhancer factor 2D) is a transcriptional activator that belongs to the MADS-box family. It binds specifically to the MEF2 element, 5'-YTA[AT](4)TAR-3', found in numerous muscle-specific, growth factor- and stress-induced genes. It controls muscle and neuronal cell differentiation and acts as a survival factor, particularly in neurons. It plays a critical role in stress-dependent cardiac growth and has been implicated in immune regulation by controlling T-cell differentiation. It plays diverse roles in the control of cell growth, survival and apoptosis via p38 MAPK signalling in muscle-specific and/or growth factor-related transcription.
