Target Full Name: Myocyte-specific enhancer factor 2A

Target Alias: MEF2; myocyte enhancer factor 2A; RSRFC4; RSRFC9; Serum response factor-like protein 1

Product Type Specific: MEF2A phosphosite-specific antibody

Antibody Code: PN622

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: T108

Protein UniProt: Q02078

Protein SigNET: MEF2A

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human MEF2A

Antibody Immunogen Sequence: SYVL(pT)PHTE(βA)C

Antibody Immunogen Description: Corresponds to amino acid residues S104 to E112. The effect of T108 phosphorylation is unclear. This is one of the major in vivo phosphorylation sites in MEF2A (≥26 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). MEF2A is known to be phosphorylated at this site in vitro by p38a MAPK (MAPK14).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus macaque | Rat | Platypus | Frog | Fruit fly.

Scientific Background: MEF2A (Myocyte-specific enhancer factor 2A) is a transcriptional activator that belongs to the MADS-Box family. As a homo- or heterodimer, itbinds specifically to the MEF2 element, 5'-YTA[AT](4)TAR-3', which is found in numerous muscle-specific genes. It is also involved in the activation of numerous growth factor- and stress-induced genes. It mediates cellular functions not only in skeletal and cardiac muscle development, but also in neuronal differentiation and survival. It functions in diverse roles in the control of cell growth, survival and apoptosis via p38 MAPK signaling in muscle-specific and/or growth factor-related transcription. Phosphorylated and sumoylated MEF2A represses transcription of NUR77 in cerebellar granule neurons, and promotes synaptic differentiation. It associates with chromatin to the ZNF16 promoter. MEF2A is also recognized for its role in neuronal connectivity and mitochondrial function, with links to oxytocin-induced effects. Isoform MEF2 and isoform MEFA are expressed only in skeletal and cardiac muscle and in the brain. Isoform RSRFC4 and isoform RSRFC9 are widely expressed. Defects in the MEF2A gene can lead to Autosomal Dominant Coronary Artery Disease 1 (ADCAD1) with myocardial infarction. This description may include information annotated by UniProt and/or Google AI.
