Antibody Immunogen Description: Corresponds to amino acid residues V151 to T159. T155 phosphorylation regulates transcription, although one report indicates inhibition, wheras another describes induction. This is one of the major in vivo phosphorylation sites in LEF1 (≥11 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). LEF1 is known to be phosphorylated at this site in vitro by NLK.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: LEF1 (Lymphoid enhancer-binding factor 1) is a nuclear transcription factor and functions as part of the TCF/LEF family of transcription factors. It is a key downstream mediator of the Wnt/beta-catenin signaling pathway, It functions in regulating cell proliferation, differentiation, and stem cell maintenance. LEF1 binds to beta-catenin, transforming it into a transcriptional activator of Wnt target genes (e.g., MYC, CCND1). PIAG antagonizes both Wnt-dependent and Wnt-independent activation by LEF1. TLE1, TLE2, TLE3 and TLE4 repress transactivation mediated by LEF1 and beta-catenin. It regulates T-cell receptor alpha enhancer function. It may acts as a positive regulator of odontoblast differentiation during mesenchymal tooth germ formation, expression is repressed during the bell stage by MSX1-mediated inhibition of beta-catenin signaling. It contains an HMG-box domain for DNA binding and an N-terminal domain for beta-catenin interaction. Some isoforms (Isoform 3) can act as transcriptional repressors by binding to co-repressors like CtBP when Wnt signaling is absent. It is primarily expressed in pre-B and T cells (lymphoid tissues) in thymus, but also in developing tissues like hair follicles and teeth.
