Antibody Immunogen Description: Corresponds to amino acid residues M235 to L242. T239 phosphorylation inhibits transcriptional activity by reducing binding of DNA. Its phosphorylation has been separately reported to both induce and inhibit FBXW7 interaction. This is one of the major in vivo phosphorylation sites in c-Jun (≥24 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Jun is known to be phosphorylated at this site in vitro by GSK3b.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: c-Jun is a transcription factor in the AP1 complex that recognizes and binds to the enhancer heptamer motif 5'-TGA[CG]TCA-3'. It is activated by c-Jun N-terminal kinases (JNKs) through phosphorylation at specific residues (Ser-63 and Ser-73). It regulates gene expression (e.g., cyclin D1), cell proliferation, and apoptosis, playing a critical role in tissue development and neuronal responses to injury. It is activated by JNK phosphorylation. It promotes activity of NR5A1 when phosphorylated by HIPK3, which leads to increased steroidogenic gene expression upon cAMP signalling pathway stimulation. It features a bZIP domain (basic leucine zipper) crucial for DNA binding and dimerization. c-Jun is the cellular homologue of the viral oncoprotein v-Jun and is often upregulated in various cancers, promoting tumorigenesis.
