Antibody Immunogen Description: Corresponds to amino acid residues V53 to L61. Effects of Y56 and pY58 phosphorylation on this translation factor are unclear. These are the major in vivo phosphorylation sites in HBS1L with 145 and 256 mass spectrometry reports, respectively in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: HBS1L (HBS1-like translational GTPase) is a protein that acts as a key ribosomal rescue factor, is crucial for the No-Go Decay (NGD) pathway to degrade faulty mRNAs and manage stalled ribosomes. As a GTPase, HBS1L forms a complex with Pelota (PELO) to recognize stalled ribosomes at the 3' end of mRNA, facilitating their disassembly and the subsequent degradation of damaged mRNA. Following mRNA extraction from stalled ribosomes by the SKI complex, the Pelota-HBS1L complex promotes recruitment of ABCE1, which drives the disassembly of stalled ribosomes, followed by degradation of damaged mRNAs as part of the NGD pathway. The HBS1L gene is located on chromosome 6q in the HBS1L-MYB intergenic region, which is a major genetic determinant of fetal hemoglobin (HbF) levels, influencing erythrocyte and platelet counts. Variations in the HBS1L-MYB intergenic region are strongly associated with the regulation of HbF levels, which is critical for modulating the severity of sickle cell disease and beta-thalassemia.
