Antibody Immunogen Description: Corresponds to amino acid residues D261 to L268. S265 phosphorylation stabilizes protein. This is the major in vivo phosphorylation site in FRA1 (≥13 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). FRA1 is known to be phosphorylated at this site in vitro by PKCt.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: FRA-1 (Fos-related antigen 1; FOSL1) is a basic leucine zipper (bZIP) transcription factor and belongs to the bZIP family, Fos subfamily. It is a key member of the AP-1 complex, and it forms heterodimers with Jun family proteins to bind to DNA. It acts by binding to TRE (TPA reaction element), CRE (cAMP-responsive element), and SRE (serum response element) on DNA. It regulates gene expression related to tumour proliferation, angiogenesis, epithelial-to-mesenchymal transition (EMT), invasion, and metastasis. Its stability is highly dependent on phosphorylation by kinases such as ERK2, Rsk1, and PKC-theta. Its expression is regulated by oncogenes like Myc and Ras. It is crucial for cancer metastasis, particularly in breast, lung, colorectal, and thyroid cancers, where it is often over-expressed. It also promotes the M2d polarization of macrophages in the tumour microenvironment, further driving cancer progression. As part of the AP-1 complex, it binds to DNA to initiate transcription of target genes. It is induced by growth factors and cytokines.
