Antibody Immunogen Description: Corresponds to amino acid residues P607 to K614. T611 phosphorylation facilitates binding histone acetyltransferase CREB binding protein (CBP) to the FoxM1 transcriptional activation domain. This is one of the minor in vivo phosphorylation sites in FOXM1 based on ≥6 high throughput mass spectrometry reports recorded in PhosphoSitePlus. FOXM1 is known to be phosphorylated at this site in vitro by CDK1 (CDC2), CDK2, CDK4, and CDK6.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: FOXM1 (Forkhead box protein M1) is a transcriptional factor in the Forkhead family. It features an N-terminal Repressor Domain (NRD), a conserved Forkhead (winged-helix) DNA Binding Domain (DBD), and a C-terminal Transactivation Domain (TAD). It promotesthe expression of cell cycle genes essential for DNA replication and mitosis (especially the G1/S and G2/M transitions). FOXM1 drives mitosis by activating genes such as Cdc25B, Aurora B kinase, and cyclin B1. In addition to control of cell proliferation, it is also involved in repair of DNA breaks participating in the DNA damage checkpoint response. It also promotes transcription of PHB2. It is essential for tissue development and regeneration but, when overexpressed (in up to 85% of certain cancers), acts as an oncogene in most human cancers. It is expressed in thymus, testis, small intestine, colon followed by ovary. It appears to be expressed only in adult organs containing proliferating/cycling cells or in response to growth factors. It is also expressed in epithelial cell lines derived from tumours, but not in resting cells. Isoform 2 is highly expressed in testis.
