Target Full Name: Forkhead box protein K1

Target Alias: MNF; Myocyte nuclear factor

Product Type Specific: FOXK1 phosphosite-specific antibody

Antibody Code: PN590

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: S441+S445

Protein UniProt: P85037

Protein SigNET: FOXK1

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human FOXK1

Antibody Immunogen Sequence: CL(pS)REG(pS)PIP

Antibody Immunogen Description: Corresponds to amino acid residues C439 to P448. The effects of S441 and S445 phosphorylation are unclear. These are two of the major in vivo phosphorylation sites in FOXK1 with ≥183 and ≥219 mass spectrometry reports, respectively, recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Dog | Rat | Mouse | Cow | Frog | Sea urchin .

Scientific Background: FOXK1 (Forkhead Box K1) is a versatile forkhead transcription factor that regulates metabolism, cell proliferation, and differentiation by acting as a context-dependent activator or repressor of target genes. It functions in metabolic reprogramming toward aerobic glycolysis (Warburg effect), promotes autophagy, and acts as an oncogene in cancers like lung, gastric, and colorectal by driving metastasis and tumour growth. FOXK1 binds to specific DNA motifs (5'-GTAAACA-3') and regulates genes involved in glycolysis (e.g., HK2, PKLR), autophagy, and muscle cell differentiation. It works with FOXK2 to promote lactate production while suppressing mitochondrial pyruvate oxidation. It acts as a downstream effector of the mTORC1 signaling pathway, translocating to the nucleus in response to energy status to regulate anabolic pathways. It also enhances Wnt/beta-catenin signaling to promote tumour progression. Elevated FOXK1 expression is linked to epithelial-to-mesenchymal transition (EMT), increased tumour cell proliferation, invasion, and angiogenesis.
