Antibody Immunogen Description: Corresponds to amino acid residues P228 to E236. T232 phosphorylation stimulates transcriptional activity. This was not identified as an in vivo phosphorylation site in Fos by high throughput mass spectrometry studies recorded in PhosphoSitePlus. Fos is known to be phosphorylated at this site in vitro by ERK1 (MAPK3), ERK2 (MAPK1), ERK5 (MAPK7), and Frk.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

The Fos family of transcription factors (including c-Fos, FosB, FosL1, FosL2) are immediate-early genes encoding leucine zipper proteins that dimerize with Jun family proteins to form the Activator Protein-1 (AP-1) complex. In the heterodimer, FOS and JUN/AP-1 basic regions each seems to interact with symmetrical DNA half sites. with TGF-beta activation, it forms a multimeric SMAD3/SMAD4/JUN/FOS complex at the AP1/SMAD-binding site to regulate TGF-beta-mediated signaling. It has a critical function in regulating the development of cells destined to form and maintain the skeleton. It plays important roles in processes like cell growth, differentiation, and neuronal plasticity, with high expression levels often associated with neural stimulation and development. In growing cells, activates phospholipid synthesis, possibly by activating CDS1 and PI4K2A. This activity requires tyrosine-dephosphorylation and association with the endoplasmic reticulum.
