Antibody Immunogen Description: Corresponds to amino acid residues T18 to P25. S22 phosphorylation inhibits transcriptional repression by ETV6, allowing for increased activation of ETV6-repressed genes. This is one of the major in vivo phosphorylation sites in ETV6 (≥149 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). ETV6 is known to be phosphorylated at this site in vitro by p38a MAPK (MAPK14).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ETV6 (Tel) is a ubquitous transcription factor that belongs to the ETS family. It functions by binding to specific DNA sequences through its C-terminal ETS domain and can dimerize using its N-terminal pointed (PNT) domain (helix-loop-helix). It is a transcriptional repressor that binds to the DNA sequence 5'-CCGGAAGT-3'. It inhibits target genes to manage cell proliferation, differentiation, and survival, particularly in blood development .It regulates hematopoiesis, vascular development, embryonic growth and malignant transformation. It possesses an N-terminal pointed (PNT) domain (helix-loop-helix) for self-association and a C-terminal DNA-binding ETS domain. It is ubiquitously expressed, with higher levels in the bone marrow, spleen, and thymus. It functions as a tumour suppressor by binding to DNA via its ETS domain and repressing target gene activity. The ETV6 gene is frequently involved in chromosomal translocations (e.g., ETV6-RUNX1 in childhood leukemia), and germline mutations that cause inherited thrombocytopenia and increase the risk of hematological malignancies.
