Antibody Immunogen Description: Corresponds to amino acid residues R279 to S289. S282 phosphorylation inhibits transcriptional activity by inhibiting DNA binding. This is one of the major in vivo phosphorylation sites in Ets-1 (≥95 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Ets-1 is known to be phosphorylated at this site in vitro by CaMK2a.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Ets-1 (C-ets-1) is a transcription factor that belongs to the ETS family. It directly controls the expression of cytokine and chemokine genes in a wide variety of different cellular contexts. It may control the differentiation, survival and proliferation of lymphoid cells. It also regulates angiogenesis through regulation of expression of genes controlling endothelial cell migration and invasion. It plays a critical role in immune system function, particularly in T-cell and B-cell development. It features a highly conserved DNA-binding ETS domain that adopts a winged helix-turn-helix structure, allowing it to bind to specific DNA sequences containing a GGAA/T core motif. It is highly expressed in lymphoid tissues, such as the thymus, spleen, and lymph nodes, as well as in endothelial cells. Isoforms c-ETS-1A and Ets-1 p27 are both detected in all fetal tissues tested, but vary with tissue type in adult tissues. It is not detected in adult brain or kidney. Its overexpression is linked to aggressive tumour characteristics and angiogenesis in cancers. Conversely, decreased expression is linked to systemic lupus erythematosus (SLE).
