Antibody Immunogen Description: Corresponds to amino acid residues A15 to G25. S19 and S22 phosphorylation represses transactivation activity in response to coactivator. Phosphorylation enhanced the sumoylation at Lys-14, and contributes to regulating GRIP-1-dependent activated transcriptional activation by ERRα1. These are two of the major in vivo phosphorylation sites in ESRRA (≥21 and ≥18 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ESRRA (ERR1) is a 45.5 kDa transcription factor of the nuclear hormone receptor family, NR3 subfamily. It is a master transcriptional regulator of cellular energy metabolism, mitochondrial biogenesis, and lipid metabolism. It is essential for fatty acid oxidation, mitochondrial function, and energy metabolism. Its deficiency results in impaired metabolism and reduced adaptation to stress. ESRRA binds to an ERR-alpha response element (ERRE) containing a single consensus half-site, 5'-TNAAGGTCA-3'. It targets the medium-chain acyl coenzyme A dehydrogenase (MCAD) response element NRRE-1, the MCAD promoter and C1 region of the lactoferrin gene promoter. It requires dimerization and the coactivator, PGC-1A, for full activity. The ERRalpha/PGC1alpha complex is a regulator of energy metabolism. It induces the expression of PERM1 in the skeletal muscle. It is frequently overexpressed in various cancers (e.g., breast, ovarian, gastric, gallbladder), where it supports tumor growth, metastasis, and drug resistance.
