Antibody Immunogen Description: Corresponds to amino acid residues G522 to R529. T526 phosphorylation inhibits ERF transcriptional repression by inducing its export from the nucleus, and regulates cell cycle progression. Phosphorylation induces transcriptional activity. by inhibition of ERF repression. This is the major in vivo phosphorylation site in ERF (≥150 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). ERF is known to be phosphorylated at this site in vitro by ERK1, and ERK2.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ERF (ETS2 repressor factor) is a ubiquitously expressed ETS-family transcription factor that functions primarily as a strong transcriptional repressor, regulating cellular proliferation, differentiation, and suppressing Ets-associated tumorigenesis. It acts as a downstream effector in the Ras/MAPK signaling pathway, where phosphorylation by ERK1/2 MAP kinases causes its nuclear export and inactivation. It is a potent transcriptional repressor that binds to the H1 element of the Ets2 promoter. It may regulate other genes involved in cellular proliferation. ERF is required for extraembryonic ectoderm differentiation, ectoplacental cone cavity closure, and chorioallantoic attachment. It may be important for regulating trophoblast stem cell differentiation. Its highest levels of expression are in testis, ovary, pancreas, and heart. Mutations in the human ERF gene are associated with diseases like Craniosynostosis 4 (CRS4). ERF suppresses ets-induced transformation, and Its repression activity is often decreased in cells transformed by activated Ha-ras and v-src.
