Antibody Immunogen Description: Corresponds to amino acid residues E163 to G172. S167 phosphorylation induces transcriptional activity and regulates cell growth, apoptosis, transcription, acetylation, activation and molecular association, including inducing interaction with SRC-3. This is one of the major in vivo phosphorylation sites in the estrogen receptor (≥5 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). ERa is known to be phosphorylated at this site in vitro by Akt1 (PKBa), Akt2 (PKBb), AurKA, CK2a1 (CSNK2A1), IKKE, p70S6K (RPS6KB1), RSK1 (RPS6KA1), and RSK2 (RPS6KA3).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

It features an N-terminal modulator domain (AF-1), a central DNA-binding domain, and a C-terminal ligand-binding domain (AF-2). It binds estrogen to regulate gene expression in reproductive and metabolic tissues, acting as a crucial driver in over 70% of breast cancers. Structurally, it is a nuclear receptor with DNA-binding and ligand-binding domains that forms dimers to influence transcription and can function as an RNA-binding protein. It regulates eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. It controls metabolism, and sexual development, largely in reproductive tissues.
