Antibody Immunogen Description: Corresponds to amino acid residues E320 to G329. S324 phosphorylation stimulates transcriptional activity by MAD2L2 and potentiates ternary complex formation. This is the major in vivo phosphorylation site in Elk-1 (≥20 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Elk1 is known to be phosphorylated at this site in vitro by ERK1 (MAPK3), ERK2 (MAPK1), and JNK2 (MAPK9).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Elk1 (ETS domain-containing protein Elk-1) is a transcription factor that belongs to the ETS family. It features an N-terminal ETS domain (DNA binding), a B-box (SRF binding), and a C-terminal activation domain (phosphorylation sites). Elk1 binds to purine-rich DNA sequences. It forms a ternary complex with SRF (serum response factor) and the ETS and SRF motifs of the serum response element (SRE) on the promoter region of immediate early genes such as FOS and IER2. It induces target gene transcription upon MAPK-signalling pathways stimulation. It is activated via phosphorylation by MAP kinases (ERK, JNK, p38) upon extracellular stimulation, allowing it to recruit co-activators like p300.
