Antibody Immunogen Description: Corresponds to amino acid residues K1266 to T1273. S1270 phosphorylation inhibits transcriptional activity by inhibiting DNA binding. This is one of the major in vivo phosphorylation sites in CUX1 (≥14 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). CUX1 is known to be phosphorylated at this site in vitro by CDK1 (CDC2), and PKACa (PRKACA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CUX1 (Homeobox protein cut-like 1) is a transcription factor that belongs to the CUT homeobox family. It features containing three Cut repeats and a homeodomain, which are essential for DNA binding. It functions in the control of neuronal differentiation in the brain, in part by regulation of dendrite development and branching, and dendritic spine formation in cortical layers II-III, and also in control of synaptogenesis. In addition, it has probably a broad role in mammalian development as a repressor of developmentally regulated gene expression. The 200 kDa CUX1 isoform acts as a repressor, whereas cleaved, shorter 110 and 75 kDa isoforms can act as transcriptional activators. It may act by preventing binding of positively-activing CCAAT factors to promoters. It is a component of nf-munr repressor, and binds to the matrix attachment regions (MARs) (5' and 3') of the immunoglobulin heavy chain enhancer. It represses T-cell receptor (TCR) beta enhancer function by binding to MARbeta, an ATC-rich DNA sequence located upstream of the TCR beta enhancer. It also binds to the TH enhancer, and may require the basic helix-loop-helix protein TCF4 as a coactivator. A CDP/CUX p110 complex plays a role in cell cycle progression, particularly at the G1/S transition. As cells progress into S phase, a fraction of CUX1 molecules is proteolytically processed into N-terminally truncated proteins of 110 kDa. While CUX1 only transiently binds to DNA and carries the CCAAT-displacement activity, CDP/Cux p110 makes a stable interaction with DNA and stimulates expression of genes such as POLA1. CUX1 acts as a tumour suppressor and is frequently mutated or deleted in myeloid malignancies (MDS/AML). However, it can also act as an oncogene in certain contexts (e.g., pancreatic or breast cancer), promoting tumor cell migration, invasion, and centrosome clustering.
