Antibody Immunogen Description: Corresponds to amino acid residues C381 to E389. The effect of T385 phosphorylation is unclear. This is one of the major in vivo phosphorylation sites in CDC5L (≥66 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). CDC5L is known to be phosphorylated at this site in vitro by ERK1 (MAPK3).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDC5L (Cell division cycle 5-like protein) is 92 kDa DNA-binding and transcription protein that belongs to the CEF1 family, and functions as a dimer. It acts as a core component of the human Prp19/CDC5L spliceosomal complex, which forms an integral part of the spliceosome and is required for activating pre-mRNA splicing. The PRP19-CDC5L complex may also play a role in the response to DNA damage (DDR). It interacts with ATR (ataxia-telangiectasia and Rad3-related) and is required for the S-phase checkpoint. It is essential for pre-mRNA splicing (second catalytic step), cell cycle progression, and DNA damage repair. CDC5L is involved in cell cycle control and has a role in pre-mRNA splicing as core component of precatalytic, catalytic and postcatalytic spliceosomal complexes. It acts as a transcription activator and has been linked to the regulation of chondrocyte differentiation. It is ubiquitously expressed in both fetal and adult tissues, including lymphocytes, T-cells, and proliferating cells. Defective signalling of CDC5L is associated with intracerebral hemorrhage and polycystic kidney disease. It is overexpressed in osteosarcoma and cervical tumours.
