Antibody Immunogen Description: Corresponds to amino acid residues A307 to G314. S310 phosphorylation inhibits transcriptional activity. This is a minor in vivo phosphorylation site in the androgen receptor (≥5 reports from high throughput mass spectrometry studies in PhosphoSitePlus). AR is known to be phosphorylated at this site in vitro by CDK1, CDK5, and CDK11B.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: AR (Androgen receptor) is a steroid hormone receptor that is a ligand-activated transcription factor. These ligands include testosterone and dihydrotestosterone. Its transcription factor activity is modulated by bound coactivator and corepressor proteins like ZBTB7A that recruits NCOR1 and NCOR2 to the androgen response elements/ARE on target genes, ansd negatively regulating androgen receptor signalling and androgen-induced cell proliferation. Transcription activation is also down-regulated by NR0B2. It is activated, but not phosphorylated, by HIPK3 and ZIPK/DAPK. Isoform 2 is mainly expressed in heart and skeletal muscle. Isoform 3 is expressed by basal and stromal cells of prostate. Isoform 3 is missing the C-terminal ligand-binding domain and constitutively regulates the transcription of a specific set of canonical AR-target genes, including PSA/KLK3 and TMPRSS2, independently of steroid hormones. Isoform 4 lacks the C-terminal ligand-binding domain and constitutively activate the transcription of another specific set of genes independently of steroid hormones. Transcriptional activity is enhanced by binding to RANBP9. The drug AIM-100 (4-amino-5,6-biaryl-furo[2,3-d]pyrimidine) suppresses TNK2-mediated phosphorylation at Tyr-269 by inhibiting the binding of the Tyr-269 phosphorylated form to androgen-responsive enhancers (AREs) and its transcriptional activity.
