Antibody Immunogen Description: Corresponds to amino acid residues A183 to Q191; in the region between the SH2 and Exo-endo_phos domains. The effect of Y187 phosphorylation is unclear. This has not been confirmed as an in vivo phosphorylation site by mass spectrometry according to PhosphoSitePlus. This phosphorylation site is also conserved in chicken.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SHIP1 (SH2 domain-containing inositol 5-phosphatase 1; INPP5D) is a phosphatidylinositol (PtdIns) phosphatase that specifically hydrolyzes the 5-phosphate of phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P3) to produce PtdIns(3,4)P2, to negatively regulate PI3K (phosphoinositide 3-kinase) pathways. It acts as a negative regulator of B-cell antigen receptor signalling, and mediates signalling from the FC-gamma-RIIB receptor (FCGR2B) as well as a negative regulator of myeloid cell proliferation/survival and chemotaxis, neutrophil migration, mast cell degranulation, immune cells homeostasis, integrin alpha-IIb/beta-3 signalling in platelets and JNK signalling in B-cells.
